Probiotic Production from Bacillus Subtilis and Its Effect on Broiler Growth Performance

Abstract

The production of probiotic from spore forming bacteria genus Bacillus subtilis bearing accession number KT427377 was carried out by shake flask method. For fermentation process 1L of (LB) fermentation medium charged in 2.5L flasks, the flask tightens with cotton wool plug and then the fermented medium was kept in autoclave for sterilization (at 121°C and 15 lbs/in 2 pressure) for a period of 15 min. The fresh prepared bacterial inoculums inoculated was cooled and put in the flasks, the process of incubation was performed at 37 0C using a rotatory shaker (at 159 rpm) for 24hrs. 

The centrifugation process, of the fermented culture broth, was performed (at 10,000 rpm) for 15min for the separation of supernatant, whereas, the pellet was washed three times by using distal water and then stored for lypholization, to get the product in pure powder. The dried/powdered permitted bacillus filled in a safe container. Colony Forming Units (CFU) were determined, using the freezed dry culture, in the cell mass of probiotic with the help of serial dilution method.

After getting the product in powder form here we observed the antibacterial activity and the effects of this probiotic on broiler weight gain, food conversion ratio (FCR), growth performance and carcass parameters were studied. For above reason, the experimental 270 day-old broiler birds were purchased from local market to perform the experiment. The experimental broiler birds arranged randomly in three experimental groups G1, G2 and G3, in each group were 90 birds having six replicates each having 15 birds in environmentally controlled shed. Group 1 was negative control birds having diet without provision of probiotic and groups G2 and G3 were positive control birds having diets containing probiotic 1% and 1.5% in the starter and finisher diet respectively, feeders and drinkers were kept for feed and water.

The feed conversion ratio (FCR) in fed on probiotic-supplemented chicks was found significantly (p<0.05higher as compare to control group. Whereas, the chicks of group 3 showed a significant (p<0.05) heavier weight gains than chicks of G2. Furthermore, the other parameters viz; feed intake, composition of meat, empty organ weights and dressing percentage was found non-significant amongst the all diets. However, supplemented groups showed significantly (P<0.05) reduced abdominal fat content. So this experiment show that the probiotic produce from bacillus subtilis accession no KT427377 has beneficial effect on broiler health, weight gain and feed efficient.
